When initiating studies with a particular E3 ligase, UBE2D3 (UbcH5c) is often selected due to the reported promiscuity of this E2. However, it should be noted that several E3 ligases (including Parkin) have been reported not to work with UBE2D3. UBE2L3 (UbcH7) has been shown to function with several E3 ligases, including E6-AP and Parkin. UBE2N/UBE2V2 functions as a heterodimeric E2 enzyme for the production of K63-linked ubiquitin chains, and it is a good choice if you know that the E3 of interest builds this linkage type. This information can be a starting point in matching E2/E3 enzyme pairs for your ligase assays, but comprehensive profiling is still sorely needed.
The E2 Selection Panel is designed to provide the most flexible and cost-effective solution to this daunting task. In a single package, four* E2 conjugation enzymes from our catalog can be selected**. These enzymes can be used as stand-alone products or in conjugation with both the E3LITE Customizable Ubiquitin Ligase Kit or the E2 Profiling Kit.
