Human SUMO2-AMC is prepared via the conjugation of the C-terminus of mature human Small Ubiquitin-like Modifier (SUMO)2 to 7-amino-4-methylcoumarin (AMC), leading to quenching of the intrinsic fluorescence of AMC. Upon incubation with desumoylases, AMC is released and an increase in fluorescence can be measured using at 460 nm (excitation at 380 nm). The high purity of LifeSensors’ SUMO2-AMC results in improved to signal:background ratios. Optimization of assay parameters and reagent requirements must be determined by the end user.
