PROTACs (PROteolysis-TArgeting Chimeras) are a modern class of small molecules designed to selectively degrade disease-causing proteins rather than merely inhibiting their activity. By harnessing the cell’s natural protein disposal system (the ubiquitin–proteasome pathway), PROTACs bring a target protein into proximity with an E3 ubiquitin ligase, triggering its tagging and subsequent degradation.
