Chloroquine diphosphate is a 4-aminoquinoline anti-malarial and anti-rheumatoid agent, also acting as an ATM activator. It inhibits autophagy in a variety of cell lines, induces cell death in breast cancer cell lines, and displays antitumor and antimetastatic activity in mouse models of breast cancer. Chloroquine diphosphate eliminates cancer stem cells via deregulation of Jak2 and DNMT1. It displays synergy when combined with the Raf inhibitor, vemurafenib, in brain tumors. It is also cell permeable and active in vivo.
