USP30 (Ubiquitin Specific Protease 30) is a deubiquitinase involved in the regulation of mitophagy. It is attached to the outer mitochondrial membrane by insertion of a transmembrane domain (residues 36-56). It removes ubiquitin from target proteins ubiquitinated by the E3 Ligase Parkin (PARK2). This form of USP30 has been engineered to increase it stability while retaining the cleavage preference for K6-linked ubiquitin chains over other chains. USP30 shows reduced efficiency to cleave phosphorylated ubiquitin chains.
