UPS9999: UPS Inhibitor Cocktail

The dynamic Ubiquitin Proteasome System (UPS) rapidly eliminates ubiquitinated proteins, making key intermediates difficult to detect. In living cells, tissue and serum, proteins tagged with ubiquitin are often degraded within seconds, and deubiquitinases (DUBs) can quickly remove ubiquitin chains. Without inhibition, ubiquitinated species will certainly be lost before analysis. LifeSensors UPS inhibitor cocktail prevents loss of ubiquitin signals. This cocktail improve reproducibility by maintaining in vivo ubiquitination states, and enables accurate detection of ubiquitinated proteins in cells, tissues, and serum. LifeSensors inhibitor cocktail also helps researchers differentiate between defects in ubiquitination and rapid proteasomal degradation.

 This cocktail is designed for use with Tandem Ubiquitin Binding Entities (TUBEs) which bind to ubiquitinated proteins with high specificity, shielding them from residual DUB activity and facilitating downstream analyses such as Western blotting, immunoprecipitation, or mass spectrometry. This approach is especially useful for identifying novel UPS substrates, mapping ubiquitination sites, and distinguishing between different ubiquitin chain linkages when linkage-specific TUBEs are used.
