UBE2W is a ubiquitin conjugating enzyme that mediates the transfer of ubiquitin from a ubiquitin-activating enzyme (E1) to a substrate protein or E3 ligase. UBE2W was originally identified as the human homolog of the Arabidopsis E2 enzyme Ubc-16. Researchers demonstrated that UBE2W contains a NLS sequence and is localized to the nucleus of HEK293 cells. UBE2W binds to the E3 ligase BRCA1 and mediates the monoubiquitylation of BRCA12. In vitro UBE2W catalyzes monoubiquitination and ‘Lys-11’-linked polyubiquitination.
