UBE2S (also known as E2-24K or E2-EPF) is an E2 conjugating enzyme that is involved in the ubiquitylation of target substrates along with E1 and E3 enzymes. It has been implicated in the ubiquitylation of anaphase-promoting complex (APC/C), promoting the elongation of K11-linked polyubiquitin chains on this substrate, after initial ubiquitylation by either UBE2C or UBE2D1. This polyubiquitin chain elongation enhances the degradation of APC/C by the proteasome. UBE2S also targets von Hippel-Lindau (VHL) for proteasome degradation, resulting in the stabilization of HIF-1a.
