MMS2 is an ubiquitin E2 variant (UEV) protein that acts in concert with the E2 enzyme Ubc13. This heterodimeric complex assembles K63-linked polyubiquitin chains for non-degradative ubiquitin signaling pathways (DNA repair, signal transduction). The UEV proteins are structurally similar to E2 enzymes; they display the same fold, yet lack the active site cysteine indicative of E2 enzymes. The Ubc13/MMS2 complex is unique in that it has the ability to form free K63-linked polyubiquitin chains in solution in the absence of an E3.
