TUBE-based Mass Spec Proteomics

Tandem Ubiquitin Binding Entities (TUBEs) are powerful reagents for the enrichment of polyubiquitylated proteins. Combining this enrichment with mass spectrometric (MS) analysis allows for identifying polyubiquitinated proteins in cell/tissue lysate. The polyubiquitinated fraction of a protein is often of low abundance and is difficult to be detected by traditional MS techniques. TUBE-based enrichment of polyubiquitylated proteins has proven key for the progress of ubiquitin proteomics. A brief overview of this technology is depicted in the following figure: 

TUBE proteomics is initiated by incubating cell/tissue lysates with TUBEs. After eluting the enriched poly-ubiquitinated fraction, it is subjected to a tryptic digest followed by MS analysis. LifeSensors provides an Excel sheet containing the list of all proteins, peptides, and ubiquitylated sites that were detected by MS as well as a quality report summarizing the results of the experiment.  



1: Identification of poly-ubiquitylated proteins in a cellular or animal model. 
2: Identification of the number and position of the polyubiquitination site(s) on the protein sequence.  
3: Monitoring the abundance and the polyubiquitination state of multiple proteins at the same time. 


1: Identify ubiquitination patterns specific to treatment with successful drug discovery candidates in order to identify biomarkers for subsequent pre-clinical and clinical studies. 
2: Follow the effect of a PROTAC drug on the abundance and polyubiquitination state of multiple endogenous proteins in the cell/tissue. 
3: Assess the specificity of a PROTAC drug for its target by looking the polyubiquitination and degradation of other proteins.
