Product Description

Recombinant SUMOstar Protease 1 specifically recognizes and cleaves the SUMOstar tag from recombinant fusion proteins. Unlike thrombin, EK, or TEV protease, which recognize short, linear sequences, SUMOstar Protease 1 recognizes the tertiary structure of SUMOstar.  As a result, SUMOstar Protease  1 never cleaves within the protein of interest. SUMOstar Protease 1 contains a polyhistidine tag at the N-terminus, allowing easy removal from the cleavage reaction by affinity chromatography.  The protease cleaves consistently over a broad range of temperature (30°C is optimal), pH [5.5 – 9.5], and ionic strength.

Benefits

  • Specifically engineered to recognize and cleave the SUMOstar fusion tag .
  • High specificity (recognizes tertiary structure) and does not cleave within the protein of interest.
  • Cleavage yields native protein with a desired N-terminus (except proline); for proteins that begin with proline, please click here.
  • Active over broad temperature (2°C to 37°C) and pH (5.5-9.5) ranges.

Unit Definition

One unit of SUMOstar Protease 1 cleaves 100 µg of a SUMOstar-MetGFP in 1h at 30°C.

Storage/Stability

Store SUMOstar Protease 1 at -80°C. Avoid repeated freeze-thaw cycles. The enzyme is stable for one year under these conditions.
