Our SUMO-based Protein and Peptide Expression Systems maximise the yield of soluble, functional proteins in E. coli, Yeast, Insect cell, and Mammalian cell cultures.  SUMO acts partially like a chaperonin and partially as an initiator of protein folding to dramatically improve the solubility of the protein of interest when expressed at high levels.  Our desumoylases efficiently and robustly remove the SUMO tag exactly at the junction of the C-terminus of SUMO and the N-terminus of your protein to release it with the N-terminal amino acid you desire (with the exception of proline).  Both SUMO and the desumoylases have hexaHis tags making their subsequent removal fast and easy.  Currently, our SUMO-tags come in three flavors: SUMOpro, SUMOpro-3, and SUMOstar.

• SUMOpro is only available for expression in bacteria as it is recognized and cleaved by endogenous eukaryotic desumoylases

• SUMOpro-3 is available for expression in bacteria and in Pichia pastoris

• SUMOstar is a version of SUMO that has been engineered to be resistant to cleavage by eukaryotic desumoylases and is available for expression in bacteria, saccharomyces, insect cells and mammalian cells.

Each system is available in a convenient kit format containing all of the necessary reagents or as individual components.
